Platelet activating factor-induced mast cell degranulation is inhibited by rupatadine, and to a lower extent by levocetirizine and desoratadine, in a mast cell line (LAD-2)

چکیده
Background Platelet activating factor (PAF) is a lipid mediator that appears to be involved in the pathophysiology of several allergic reactions such as anaphylaxis and potentially urticaria and allergic rhinitis. The role of rupatadine, a drug with dual antihistamine and anti-PAF effect, in mast cell (MC) degranulation is not known. The objective of this study was to investigate the expression of PAF receptors and the effect of rupatadine on PAFinduced MC degranulation compared with other second generation antihistamines (desloratadine, levocetizine) and a pure specific PAF inhibitor in a human mast cell line (LAD-2).

Effects of Rupatadine on Platelet- Activating Factor-Induced Human Mast Cell Degranulation Compared With Desloratadine and Levocetirizine (The MASPAF Study).
BACKGROUND AND OBJECTIVE Platelet-activating factor (PAF) is a lipid mediator involved in the pathophysiology of several allergic diseases, for example, in the amplification of mast cell (MC) activation in anaphylaxis. Rupatadine is an antihistamine with a demonstrated anti-PAF effect, although its capacity to inhibit PAF-induced MC degranulation has not been fully evaluated. Objectives: To comp...

Objective(s) Aristolochia indica has been widely used in the traditional medicine for the treatment of a variety of diseases. In the present study different extracts of roots of A. indica were evaluated for their antiinflammatory, antipruritic and mast cell stabilizing activity. Materials and Methods Anti-inflammatory activity was performed by compound 48/80 induced rat paw edema model and ...
متن کاملAmarogentin Displays Immunomodulatory Effects in Human Mast Cells and Keratinocytes
Keratinocytes express the bitter taste receptors TAS2R1 and TAS2R38. Amarogentin as an agonist for TAS2R1 and other TAS2Rs promotes keratinocyte differentiation. Similarly, mast cells are known to express bitter taste receptors. The aim of this study was to assess whether bitter compounds display immunomodulatory effects on these immunocompetent cells in the skin, so that they might be a target...
متن کاملEvaluation of nasal symptoms induced by platelet activating factor, after nasal challenge in both healthy and allergic rhinitis subjects pretreated with rupatadine, levocetirizine or placebo in a cross-over study design
BACKGROUND Platelet-activating factor (PAF) is produced by most inflammatory cells and it is involved in inflammatory and allergic reactions. We aimed to assess the anti-PAF effects of rupatadine and levocetirizine in the upper airways. FINDINGS Healthy volunteers (HV, N = 10) and seasonal allergic rhinitis (SAR, N = 10) asymptomatic patients were treated out of the pollen season with either ...
متن کاملProteinase-activated receptor-2-activating peptides induce leukocyte rolling, adhesion, and extravasation in vivo.
Proteinase-activated receptor 2 (PAR2) has been suggested to play a role in inflammatory reactions. Because leukocyte-endothelial cell interactions are critical events during inflammatory reactions, and because PAR2 is expressed both on endothelium and leukocytes, we have examined the effects of PAR2-activating peptides (PAR2-APs) on leukocyte rolling and adhesion in mesenteric venules and on l...
